1 Reactivity: Does the GM/GMN test kit react to analytes other than galactomannan ?
No. Only galactomannan is the active substance of ELISA.
The product is an indirect competition microplate assay for the detection of Aspergillus galactomannan antigen using specific antibodies. Transferring the pre-treated mixtures of the human serum sample or BAL fluid and the anti-galactomannan antibodies into the microplate which have been coated with the galactomannan antigen. After the microplate was incubated and washed, the Anti-mouse IgG conjugated with HRP were added. Following a second incubation and washing, Chromogen TMB Solution was added to form a color reaction. The absorbance value was determined by a microplate reader at 450nm wavelength. The absorbance value has a negative correlation with the content of galactomannan. Then the concentration of the galactomannan in the human serum or BAL fluid can be calculated from the standard curve and the quantitative detection can be performed.

5 Assay: Which instrument is suitable for the GM/GMN assay?

6 Assay: What software is required to run the GM/GMN assay?

9 Interpretation: Does a positive value always mean that the patient has an aspergillus infection?
No. The entire clinical context must be considered. Exclude the contamination, the positive result indicate a possible infection.
10 Interpretation: How often should a patient be tested?
Patients should be tested 2-3 times per week. Serum or BAL fluid galactomannan levels are dynamic.
